Characterizing microorganisms according to different criteria is useful when investigating sources of microbiological contamination in the pharmaceutical industry. The aim of this study was to characterize 38 Acinetobacter baumannii complex strains isolated from a biopharmaceutical industry by 16S rRNA sequencing, matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF/MS), multilocus sequence typing (MLST), antimicrobial susceptibility profile, biofilm formation, and sensibility to disinfectants. Thirty-three (86.9%) strains were identified by 16S rRNA gene sequencing as A. seifertii/pitti/nosocomialis/lactucae, four (10.5%) as A. baumannii, and one (2.6%) as A. vivianii/courvalini. MALDI-TOF/MS did not identify one strain, and incorrectly identified 30/37 (81.1%) strains as A. baumannii. Strains were assigned to 12 different STs, of which nine were newly defined in this study (STs 2091-2099). Twenty-six (68.4%) strains showed resistance to amikacin and gentamicin. Thirty-three (86.8%) strains were classified as moderately or strongly adherent on polystyrene. Alcohol 70%/15 min and quaternary ammonium 0.08%/20 min were not able to eliminate the biofilm formed, but sodium hypochlorite 0.1%/15 min was efficient. In conclusion, improved methods are needed to improve the identification of Acinetobacter strains in pharmaceutical industries. This organism is of particular concern as it forms recalcitrant biofilms, leading to persistence in the manufacturing environment and increased risk of product contamination.

The Hen's Egg Test - Chorioallantoic Membrane (HET-CAM) is a valid alternative method used to assess the potential for eye irritation from chemicals. This method is the only one that mimics the conjunctivae of the eye and aims to semi-quantitatively evaluate the irritant potential of a chemical on the chorioallantoic membrane surrounding the chicken embryo in egg by observing the irritation effects on the membrane immediately after the pure or diluted chemical is applied. The purpose of this study was to compare the different protocols of the HET-CAM, the French and German protocols, by evaluating the eye irritation potential of surfactants. The comparison led to the optimization of the French protocol, generating an adapted one, to reduce subjectivity in the test evaluation, ensuring more accurate results and greater quality control. The comparison showed that there are no significant differences between the results obtained in the French and German protocols. HET-CAM is known to overestimate the results and to be able to accurately identify non-irritant products and it is a great candidate to be part of a Bottom-up test strategy. It also can be used in a battery of tests to completely replace rabbits.

Corynebacterium ulcerans has been increasingly isolated as an emerging zoonotic agent of diphtheria and other infections from companion animals. Since pets are able to act as symptomless carriers, it is also essential to identify virulence potential for humans of these isolates. In this work the ability of C. ulcerans to bind to fibrinogen (Fbg), fibronectin (Fn) and Type I collagen as well the genetic relationship among strains isolated from human and asymptomatic dogs in Rio de Janeiro (Brazil) were analyzed. Five pulsed-field gel electrophoresis (PFGE) profiles were demonstrated (I, II, III, IV and V). In addition, the IV and V profiles exhibiting ≥85 % similarity were expressed by the BR-AD41 and BR-AD61 strains from companion dogs living in the same neighborhood. Independent of the PFGE-types, human and dog isolates showed affinity to Fbg, Fn and collagen. Heterogeneity of PFGE profiles indicated endemicity of C. ulcerans in the Rio de Janeiro metropolitan area. Differences in the expression of adhesins to the human extracellular matrix may contribute to variations in the virulence and zoonotic potential of C. ulcerans strains.

OBJECTIVE: This study presents the results of a research that aimed to describe the point of view of the population in terms of the prevalence of Adverse Reactions (AR) and information about the surveillance system. METHODS: A structured questionnaire was applied to a random sample of 200 people from the administrative staff of the Municipal Guard of Rio de Janeiro. RESULTS: 38% of the participants declared AR to some cosmetic product used in the past two years. CONCLUSION: To our knowledge, this is an unpublished study in Brazil, which presents results regarding the estimated prevalence of AR similarly to international studies. .


INTRODUÇÃO: Este trabalho integra um estudo que investiga a qualidade dos produtos cosméticos e avalia o sistema de cosmetovigilância. OBJETIVO: Ele apresenta os resultados de um inquérito cujo objetivo foi descrever o ponto de vista populacional em termos da prevalência de Reações Adversas (RA) e informações sobre o sistema de vigilância. MÉTODOS: Um questionário estruturado foi aplicado a uma amostra aleatória de 200 funcionários administrativos da Guarda Municipal do Rio de Janeiro. RESULTADOS: 38% dos participantes declararam RA a algum produto cosmético utilizado nos últimos dois anos. CONCLUSÃO: Pelo nosso conhecimento, este é um estudo inédito no Brasil que apresenta resultados em relação à prevalência de RA estimados de forma semelhante aos das pesquisas internacionais. .

Recent changes in Brazilian legislation for commercial disinfectants have been published due to the recent epidemic of nosocomial infections caused by rapidly growing mycobacteria (RGM) in many states of Brazil over the last 8years. One of these documents requires that all the manufacturers provide evidence of efficacy of sterilizing and disinfectant products, used for semi critical medical devices, against the Mycobacterium bovis BCG Moreau and Mycobacterium abscessus subsp. bolletii INCQS 00594 strains by using the Confirmative in vitro Test for Determining Tuberculocidal Activity of Disinfectants recommended by the Association of Official Analytical Chemists. These changes have caused additional costs and increased problems for importation of enrichment products at national laboratories where disinfectant efficacy assay service is performed. Middlebrook ADC Enrichment (ADC) is provided by a unique manufacturer and used in the official protocol. The aim of the present study was to evaluate an alternative in house low-cost enrichment composed of fetal bovine serum and glucose (FBSG) with ADC for performance of disinfectant efficacy assay against mycobacteria. After obtaining the growth curves for M. abscessus ATCC 19977, M. abscessus subsp. bolletii INCQS 00594, Mycobacterium chelonae ATCC 35752, and Mycobacterium fortuitum ATCC 6841 by using ADC enrichment and FBSG in Kirchners and 7H9 culture media. Through statistical analysis via the Kruskal-Wallis test on the evaluation of microorganism growth rate, it was observed that there was no inhibition of RGM growth by any of the enrichments used. These results suggest that low-cost enrichment FBSG may be used as a potential substitute of ADC for composition of media for mycobacterial growth, including in disinfectant tests.

Intoxicações não-intencionais ocorrem principalmente na infância devido à ingestão de produtos de uso doméstico. Um fator determinante é a falta de conhecimento sobre a toxicidade destes produtos. Um estudo aleatório de 158 rótulos de produtos de limpeza foi realizado no Instituto Nacional de Controle de Qualidade em Saúde - Brasil. Advertência sobre os riscos à saúde, orientações para os primeiros-socorros em caso de envenenamento e instruções de armazenamento foram avaliadas para verificar a qualidade das informações fornecidas ao consumidor sobre os riscos inerentes ao uso destes produtos. Do total de produtos avaliados, 75 por cento foram considerados inadequados porque não forneciam todas as informações necessárias para prevenir danos à saúde decorrentes do seu uso. Instruções para os primeiros-socorros no caso de inalação foram omitidas em mais de 50 por cento dos rótulos estudados e 47 por cento não recomendavam levar o rótulo para o médico em caso de acidente. Além disto, os rótulos não forneciam outras importantes advertências como ler antes de usar e manter no frasco original. Os resultados indicam que a rotulagem dos produtos de limpeza de uso domiciliar não fornece todas as informações para garantir a segurança da população.
